Hey, I'm not familiar with the character you wanted to draw, so I can only give general advise. But if you tried to step from anime reference to more realistic depiction, that will naturally be very hard, as you have to reverse engineer the simplification and invent details in the face that many anime stylizations omit. For why the character feels masculine, this is based in a construction error of the base face shape. You've drawn the head at a tricky downtilt angle, so don't be discoured, it's easy to mess up here. But no matter how you tilt and angle the face, all of its features need to follow parallel lines on the front plane (except if we add extreme foreshortening). Your features are drifting a little bit, and probably due to the angle, you set the jaw line very low and also at a different angle, which makes it look very bulky. A bulky jaw is often connotated with masculinity. In the same line of construction, you also gave her a very large ear, which will compound this effect of how massive the presence of the jaw and the bulk of the head feels. For more femininity, you could also try to place the cheekbones even higher. For the lips, you already are on the right path in not outlining them completely, but often it is a better idea to outline as little as possible. And if you emphasize the S-curves that we have typically where the lips meet in the middle, following the basic volumes that make the lips, and give them a bit of line weight and shading, we can achieve much more feminine looking lips. The fact that apart from the lips the portrait does not really use shading makes their shape also stick out more.
